MicroTime 200: Time-resolved Single Molecule Fluorescence Microscopy and what can be done….

The aim of the talk is to introduce MicroTime 200 as the all-in-one platform for time-resolved microscopy with single molecule sensitivity. Modern time-resolved  spectroscopic and microscopic measurements with single-molecule sensitivity allows monitoring of fluorescence dynamic from sub-nanosecond  to fluorescence fluctuations over seconds. Here we will explain the underlying data acquisition technique (Time-Tagged Time-Resolved (TTTR) recording)  which allows to store individually timing as well as spectral and polarization information for every photon, based on a multichannel detection scheme. Having all available information with picosecond time resolution at hand, the recorded data can be analyzed in a manifold manner, employing complementary analysis schemes like fluorescence  lifetime imaging (FLIM), Fluorescence Correlation Spectroscopy (FCS) and anisotropy imaging. Emphasis will be given on how time resolved measurements could be  employed for monitoring Förster Resonance Transfer (FRET) related assays. Finally a very brief overview of single -molecule studies will be presented with a couple of examples illustrating the single molecule sensitivity of the MicroTIme200.  More advanced analysis schemes like Fluorescence Lifetime-filtered Correlation Spectroscopy (FLCS), anti-bunching and pattern matching analysis could be presented only if time allows.

Based on the underlying technologie there will be examples shown for Biologie, Material Science, fast lifetime Imaging and super resolution.

Short bio:
Uwe Ortmann currently works as the Head of Global Sales at PicoQuant GmbH in Berlin, Germany.

He conducted his studies in Physics at the University in Kiel, Germany, and finished his thesis in 1993. He worked as product manager at BiosQuant GmbH, Berlin, Germany till 1996 and as development engineer at Edinburgh Instruments, Edinburgh, UK till 1998. He becomes than head of system development at PicoQuant GmbH and focusses fully on sales and marketing since 2006.

In his whole carrier he focusses on laser spectroscopy especially time-resolved studies, spectrometer design and finally laser scanning microscopy and single molecule detection.

Uwe Ortmann is also a senior member of the SPIE society.
